First come, first served
New applications are dealt with in the order TfL receives them. The quickest way through is a complete application: every document, the fee paid, and your DBS check and medical done, because TfL does not process an incomplete one.
If someone offers a fast track
TfL says it does not offer a fast-track service and that anyone who does is acting fraudulently. Report them to TfL's taxi and private hire team. No Knowledge school can speed up an application or an appearance either.
